Cell proliferation inhibition was estimated by MTT assay. The occurrence of apoptosis was evaluated by EtBr/AO double staining technique, cell cycle analyses and detection of apoptotic cells by annexin V- FITC and propodium iodide (PI). Differentiation of the cells was determined by NBT reduction assay and the expression of specific cell surface markers such as CD-14 and CD-11b, were analysed by flow cytometry.

When growth factor of mitochondria decreases it will leads to BCL 2 inactivation.BCL 2 gene are replaced by BAK/BAX gene .This gene are pro-apoptic gene when BCL 2 gene (antiapoptic) inactivate it will activate BAK/BAX gene.It will increase mitochondrial permeability in which cytochrome c move in cytoplasm.This activate APAF-1 which will activate caspase 9 this leads activation of caspase 3/6/7 .This whole caspase increase activation protease and endonuclease which leads to cell death.
