CHE 107- Forensic Chemistry I - Forensic DNA Lab Exercise Please answer the questions below in the spaces provided and upload this document onto blackboard. CASE SCENARIO: A woman is jogging in the park, when she is approached by a man and forced off the jogging path into a wooded area. The woman is raped by the man, who then flees on foot. The police arrive and receive a description of the rapist from the victim. Two suspects, both fitting the description of the rapist are arrested not far from the park. Both suspects are placed in a police line up, but the victim is unable to identify anyone in the line up as her attacker. The victim’s clothing and the clothing of both suspects is collected and sent to the crime laboratory for examination and DNA testing. A semen stain is found on the victim’s underwear. The victim and both suspects also gave a buccal (oral) swab to use as a DNA reference. 1. The semen stain found on the victim’s underwear was subjected to short tandem repeat (STR) DNA testing via the polymerase chain reaction (PCR). The DNA results are displayed in the electropherogram below. Interpret the DNA results by completing the table of DNA profile results. Fill in the DNA profile at each tested location for the underwear semen stain. sample D8S1179 D21S11 D3S1358 TH01 D13S317 D19S433 vWA TPOX AMEL D5S818 FGA underwear semen stain 15, 16 29,31 15,16,17 6,10 8,11 14,15,16 15,16,17 11,12 X,y 9,11 19,20,21 victim 16, 17 29, 31 15, 16 6, 10 8, 11 15 16, 17 11, 12 X 10 20, 21 suspect #1 15, 16 29 16, 17 6, 7 8, 12 14, 16 15, 17 8, 12 X, Y 9, 11 19, 22 suspect #2 13, 15 30 14, 16 8, 9 11, 12 17 16, 18 11, 12 X, Y 9, 11 19, 20 2. How many different STR loci (locations) were tested for all of the samples? 11 different STR loci were tested for all samples. 3. What locus gives the sex of the DNA that was detected? Was male or female DNA detected on the underwear? Male DNA was detected on the underwear. 4. Which STR locus gave 16 and 17 alleles as a result for one of the suspects? Which suspect has that result? Suspect #1 has the 16 and 17 alleles at the D8S1179 locus. 5. What alleles were detected at the TH01 locus for the underwear semen stain? What alleles were detected at the D21S11 locus for the underwear semen stain? 1. TH01 locus for the underwear semen stain: 6, 10
2. D21S11 locus for the underwear semen stain: 29, 31 6. Compare the DNA profile of the underwear semen stain to the DNA profiles generated from the reference samples of the victim, suspect #1, and suspect #2. Who is the likely contributor of the semen? The likely contributor of the semen is suspect #1 as their DNA profile matches several alleles found in the semen stain 7. Who can be excluded as a contributor of the semen? How did you come to that conclusion? Suspect #2 can be excluded as a contributor of the semen as none of their alleles match the semen stain. 8. A semen stain is also found on a pair of pants from one of the suspects. A DNA mixture consisting of DNA from two different people is detected. The DNA mixture results are in the table below. How many DNA alleles were obtained at the D3S1358 locus? How many DNA alleles were obtained at the FGA locus? D3S1358 locus: 3 DNA alleles obtained in the mixture. FGA locus: 4 DNA alleles obtained in the mixture 9. Compare the mixture to the reference samples of the victim, suspect #1, and suspect #2. Who are the likely contributors? Likely contributors to the DNA mixture found on the pants are suspect #1 and the victim, as their alleles match some of those found in the mixture. 10 . Who can be excluded from the mixture? How did you come to that conclusion? Suspect #2 can be excluded from the mixture as none of their alleles match the ones found in the DNA mixture on the pants.
