
Interpretation: Two major differences between TLC and column chromatography should be stated.
Concept introduction: Thin Layer Chromatography is a sensitive chromatographic technique which is used to analyze small quantities of the samples by solid-liquid adsorption method. Rectangular glass plate is coated with an adsorbent; sample to be analyzed is run on the plate in the developing chamber with the mobile phase to give spots of the analyzed sample.
Column chromatography is a large scale solid-liquid adsorption chromatographic technique that can separate multigram amounts of mixtures. This is a preparative version of thin layer chromatography that permits separation and isolation of products by collection of eluted fractions.
