To determine: The three types of DNA sequence that are required to ensure proper replication and propagation of YAC in a yeast cell.
Introduction: DNA cloning is a technique which is used to make genetically identical copies of a gene or segment of DNA. Recombinant DNA technology allows cloning of DNA to make recombinants. Vector such as plasmid used for cloning purposes. The desired gene can be inserted into the bacterium (host cell).
To clone large pieces of DNA in yeast cells, yeast artificial chromosomes (YACs) are required. It can generate unlimited numbers of DNA copies. Cloning genomic DNA into YAC is useful to analyze very long DNA sequence such as human genes.
