Chapter 17, Problem 1E
Summary Introduction

To explain:  Which round of the citric acid cycle will release labeled CO2, if the carbonyl carbon of acetyl CoA is labeled with 14C.

Concept introduction: The pyruvate produced from the glycolysis can enter the citric acid cycle as acetyl CoA and splits into CO2. If the carbonyl carbon of acetyl CoA is labeled, it becomes C4 of the succinyl-CoA.

Explanation of Solution

Explanation:

Citric acid cycle is central to the metabolism. Acetyl CoA enters the citric acid cycle and forms citrate on reaction with oxaloacetate. The labeled carbonyl carbon of the acetyl CoA becomes C4 of the succinyl-CoA. The CO2 produced in the first round is from the oxaloacetate carbon. The carbon of acetyl CoA is not produced as CO2 in the first round. When the labeled succinyl-CoA enters the second round, the labeled carbon is produced as CO2 by the action of isocitrate dehydrogenase and the ketoglutarate dehydrogenase enzymes.
