Inside the cell under invivo conditions the protein synthesis is carried out by complex machinery involving ribosomes, mRNA, t-RNA, and aminoacyl tRNA synthatases. it also include several other proteins which ensure the fidelity of the process. SPPS means "solid phase peptide synthesis" which was invented by merrifield group in 1963. SPPS involves the chemical reactions carried out under invitro conditions.
Advantages of invivo protein synthesis :
1. No chemicals are required except for the growth of cells.
2. Desired proteins can be synthesized invivo using recombinant DNA technology.
3. Highly efficient and it is error free. Protein synthesized under invivo conditions does not include errors in their sequences.
4. Multiple desired protein can be synthesized inside the cells using RDT.
Disadvantages of invivo protein synthesis :
1. proteins can not be synthesized in large amount in a shot time period.
2. Protein is synthesized in highly impure form.
3. Protein purification is an tedious task which take a lot of time to purify the impure protein.
4. The process can not be automated.
5. Unnatural amino acids can not be incorporated into peptides or proteins.
