A titration of amino acids is a process in which base added gradually to neutralize the acids, the functional groups of amino acids are neutralized from low to high pKa values. At low pH, net charge on all amino acids will be positive. At high pH, charge will be negative. The pH value at which no net charge on the amino acids is called as pI (isoelectric point). To find the pI value of any amino acids, we must find out the pH range where the isoelectric structure predominates. Average of the pKa values gives the pI of that amino acids.
Titration curve is graphical representation of the pH values of amino acids when sequential addition of base is performed. It s basically graph of base concentration Vs pH of solution. Generally titration curve is same for all amino acids. As we know alpha carbon of amino acids contains carboxyl, amine, side chain and H atom. Amino Acids are weak Polyprotic Acids. It is present as zwitterions at neutral pH and can be titrated with both acid/base like an amphoteric molecules. The titration curve contains equivalence point - it is basically the pH at which amino acids shows maximum buffering capacity and graph is most steep at this point.
