Uncouplers such as DNP decreases the formation of high energy phosphate and stimulate oxygen consumption. The mitochondrial transport of electron is tightly coupled with oxidative phosphorylation, that means oxidation and phosphorylation acts simultaneously. Uncouplers delink the electron transport chain from oxidative phosphorylation. They also, increase inner mitochondrial membrane permeability to proton. Therefore, uncouplers allow ETC but block establishment of proton gradient. Thus, ATP synthesis doesn't occur and energy is released as heat.
