Procedure:
Preparation of precipitating solution:
1. Place 1 spoonful of NaCl and 5 ml of liquid soap in 150 ml beaker.
2. Fill half full with hot water and mix. This is the precipitating solution.
3. In a separate 250 ml beaker, place slice banana and half fill with distilled water.
4. Add 5 tablespoon of precipitating solution and blend using blender.
5. Filter in a 250 ml beaker using cheesecloth.
6. Fill the beaker cold isopropyl alcohol
7. Let it stand and collect the fibrous mass that appears.
8. This is the DNA.
HYDROLYSIS OF DNA
A. To the fibrous mass, add 10 ml of 10% H2SO4 in a beaker. Boil gently or 5 minutes. Perform
the following test:
1. Inorganic phosphate – to 1 ml NHẠOH, add 1 ml filtrate. Acidify by placing a small piece
of blue litmus paper using 10% HNO; until the litmus paper changes to red. Then add 2 ml
ammonium molybdate. Boil the solution and allow to stand for a few minutes. Note color of
ppt.
2. Purines - to 2 ml filtrate add 3 ml of 3M NH,OH. Add 2-3 drops of 0.1M AGNO3
solution to it. Note color of precipitate.
3. Deoxyribose – to 1 ml filtrate add 1 ml of diphenylamine reagent. Heat for 10 mins in a
boiling water bath. Cool and note color produced.
