On the basis of gram staining, we can differentiate between two types of bacteria, one with thick cell wall and one with thinner one.
The procedure of gram stain is as follows:
- First step is staining with crystal violet stain which is chemically hexamethyl-para-rosaniline chloride.
- Then a decoloriser, ethyl alcohol or acetone is added. In gram positive bacteria peptidoglycan layer of cell wall is thick so, gram stain is taken and is not decolorised but in gram negative bacteria peptidoglycan layer of cell wall is thin, so it is decolorised.
- Afterwards, a counter stain usually safranin is added, that gives orangish to pink color to gram negative bacteria while gram positive remains purple.
