Bring a sponge or dishcloth used in the kitchen from home/residence.
Cut the sponge/dishcloth into sections no bigger than 10cm².
Submerge the sponge/dishcloth in a plastic bag containing 50 ml peptone broth to
extract bacteria from the item. Seal the bag.
Knead the sponge/dishcloth in the broth for 3 minutes.
With sterile forceps remove the sponge/dishcloth and place the item into a sterile
plastic bag. Seal the bag.
Wring the liquid from the sponge or dishcloth (in the bag) using your hands.
Dilute the extract and plate onto an LB and MacConkey agar plate (dilution plating
method, refer to section below).
After incubation at 25°C for 7 days, count the number of bacterial colonies and
determine the CFU/ml.
Dilution plate method:
Take six Eppendorf tubes and pipette exactly 900 µl of Ringer's solution into each.
Label the tubes clearly from 10-¹ to 10-6.
Pipette 100 μl of the extract into tube no 1 (10-1 dilution). Mix well.
Pipette 100 μl of the 10-1 dilution into the next tube and mix (10-2 dilution).
Repeat up to the 10-6 dilution.
Label four petri dishes 10-2, 10³, 104 and 10-5. Now pipette 100 μl of each dilution into
the corresponding petri dish.
Using a sterile glass rod spread the bacterial suspension over the surface of the agar.
