Osmosis Graphing Challenge Background Dialysis tubing was used to make bags, or artificial cells with different concentrations of sucrose in them. The dialysis tubing used in this experiment was made of cellulose, which is the same polysaccharide that plant cell walls are made of. This cellulose does not dissolve in water, and has pores that water can pass through, but that a disaccharide, like sucrose or table sugar, cannot pass through. The bags were placed in beakers with 1 % and 10% sucrose solutions to see how water would cross the membrane due to osmosis. Remember that osmosis is the diffusion of water across a semipermeable membrane. We weighed the bags on a balance or scale every 15 minutes to see if the mass of these bags changed over time. The bags in their beakers looked like this: This is the concentration of sucrose solution in each bag, Bag A, Bag B, Bag C, and Bag D, and the concentration of the solution in which the bag was placed: Bag A contained 1% sucrose and was placed in a solution of 10% sucrose. Bag B contained 1% sucrose and was placed in a solution of 1% sucrose. Bag C contained 10% sucrose and was placed in a solution of 1% sucrose. Bag D contained 20% sucrose and was placed in a solution of 1% sucrose. Question 1: Which of these bags were in an isotonic solution? Question 2: Which of these bags were in a hypotonic solution?

Question 3: Which of these bags were in a hypertonic solution? Data: The following table gives the weight of each bag in solution at 15 minute intervals: Bag Initial Wt. (g) 15 min. Wt. (g) 30 min. There are many sites with printable graph paper. Here is one: https://print-graph-paper.com/details/5mm . In Lab Check 2, I will ask you to upload a copy of the graph you make. Therefore you will need to make sure that you graph the total weight of each bag (not the change in weight), and that you graph the mass of all four bags on the same graph. You will use different colors or different types of lines (dotted vs. dashed vs. solid vs. dot and dash) and indicate which line belongs to which bag on a key or legend next to your graph. Your graph should have a descriptive title at the top and the axes (x and y) should be labeled with the variable and the units (minutes, grams, etc.). The graph should be sized to take up most of the graph paper, and the squares indicating the amount of time or weight should be consistent so that the graph portrays the data accurately. The values for the independent variable will be placed on the x axis of your graph. Question 5: What is the dependent variable for this experiment (what variable is dependent on the treatment)? The values for the dependent variable will be placed on the y axis of your graph. Question 6: A concentration gradient for water must be present in cells for osmosis to occur. Which bag represented the steepest concentration gradient relative to its surrounding environment?

Question 7: The steepest gradient should result in the highest rate of diffusion. Refer to your graph. Did the greatest changes in weight occur in cells with the steepest concentration gradients? Why or why not?
