N AME : C HEEK C ELL DNA-L AB R EPORT S HEET Exercise 1 – Isolating Cheek Cell DNA Analysis of Results Appearance of Solution What is happening to the cells/DNA? After lysis buffer Cloudy light blue solution After adding the lysis buffer there was a separation between the buffer and my DNA at 3mL. After adding the Protease solution and inverting it 5x’s, mixture was a pale cloudy blue throughout. After alcohol added Solution turned cloudy After adding alcohol there was a noticeable separation of the DNA from the alcohol at 6mL. While observing the solution a very thin layer of white formed at the bottom of the test tube. After alcohol mixed Solution turned to a light cloudy color at the bottom, clear solution at the top After mixing observed separation between 5.5 and 6mL and another slight separation of the DNA at 2mL. From 5.5 to 10mL solution was almost clear and from 2mL to the bottom of the test tube solution was a light cloudy color. I observed at

the bottom of the test tube there was a white stringy substance with small white particles floating in the solution. Questions: 1. Why is cell lysis the first step in isolating DNA from your cells? The lysis buffer aids in the chemical breakdown of the cell membranes. 2. Lysis buffer typically contains detergent. Why would this help to lyse the cells? The dish soap in the lysis buffer is responsible for dissolving the hydrophobic (oil-based) molecules. 3. Decribe the appearance of your DNA. Is its appearance consistent with what you know about the structure of DNA? Why or why not? When observing the appearance of my DNA it looked like a stringy white small blob. My DNA does not look like a double helix, because the double helix is microscopic. It looked like a stingy clump of white mucus 4. If you wanted to isolate a copy of the gene that codes for a protein found in the brain, could that gene be located in cheek cells? Explain your reasoning. I would be able to isolate a copy of a gene that is found in the brain by the cheek cells, because all cells in our body have the same genetic composition.

5. Meat tenderizer is often used to tenderize tough pieces of meat, like steak. Knowing that steak is made of protein-rich muscle tissue from cows, can you think of an Explanation for how meat tenderizer works? Meat tenderizer contains bromelain which is protease. This enzyme breaks down the protein molecules which results in the meat becoming tenderer.
