Chromosomes' centromeres are narrow regions that hold two sister chromatids together. Additionally, it serves as the place where microtubules are attached. Sister chromatids separate as a result of the centromere connection breaking during anaphase. Failure to do so prevents the chromatids from being separated.

Chromosome alignment on the metaphase plate requires the attachment of kinetochores to microtubules. Before they can line up on the metaphase plate, chromosomes must first undergo several rounds of shuffling.

For chromosomes to move towards the centromere's minus ends, motor proteins are necessary. Plus-end-directed motor proteins work to prevent this movement. The correct chromosome alignment at the metaphase plate is the result of a balance between these two movements.

Special proteins called kinetochore are found attached to the centromere. Every chromatid has two kinetochores. So, to separate the sister chromatids, microtubules attach to kinetochores from both poles. The tension created when microtubules from the same pole attach to kinetochores causes kinetochores to separate.
