PART B Re-watch the video and explain why the following steps are necessary: • Why use tweezers to put disks on the plate? • Why sterilize tweezers after each disk is put on the agar? Why spread the bacteria out all over the agar plate? • • Why put a disk with NO antibiotic on each agar plate? Why put a disk with newly purchased, fresh antibiotic on each plate? • Why put so many disks on each plate? • Why wait 16-20 hours after you have put the bacteria and antibiotic disks on the plate?

• Record the steps that the laboratory technician follows in the video.
• Highlight, by colour or bolding, the steps that ensure the experiment is sterile.
Materials
You will need: 6 millimeters filter paper disks, sterile Petri dishes, agar plates and no added
antibiotics, sensitive and resistant bacterial cultures, an antibiotic to test and antibiotics control
Steps
1. Pick colony and grow overnight
2. Dilute culture and grow to OD = 0.5
3. Prepare antibiotic stock
4. Add antibiotic to each disc
5. Dry discs at room temperature
6. Spread bacteria evenly on agar plate
Place discs in center of sectioned areas
7.
8. Incubate plates 16-20h and measure zones
