A. Bacteriological analysis of water: Most Probable Number (MPN) technique
1. Students should work in groups of 3 (side of a bench) and use ONE water sample
provided by them.
2. Using the diagram below as a guide, aseptically pipette 10 ml of the sample into each of
three* 10 ml double strength lactose Durham tubes; pipette 1 ml of the sample into
each of three 9 ml single strength lactose Durham tubes and finally, pipette 0.1 ml of the
sample into each of three 9 ml single strength Durham tubes.
3. Gently shake all tubes and incubate at 37°C for 48 hrs.

*Normally 5 Durham tubes are inoculated but this exercise is modified to three in the interest of economy.
4. Record the number
after 48 hrs of continuous incubation. Consult Table 1 and determine the MPN for your
tubes
each of the three dilutions of the sample that show gas
sample.
5. For each presumptive positive tube, streak an EMB plate for isolated colonies.
6. Record the Completed test MPN results. (Note that we are skipping the confirmed test).
7. Observe the demonstrations of the completed tests of the MPN. Be able to state how
and why each test is performed.

2. Why are coliforms and in particular, E. coli, "ideal indicators" of water
contamination?
