Proteins exist in four levels of molecular conformations in a human body: primary, secondary, tertiary and quaternary. The linear chain of amino acids linked together by peptide bond forms the primary structure. The secondary structure is formed by hydrogen bonding between carboxyl group of first amino acid with amino group of fourth amino acids in the polypeptide backbone. This creates a spiral structure forming an alpha-helix or beta-pleated sheet. The secondary structure further folds to form a three-dimensional structure called the tertiary structure. A protein becomes functionally active only at this stage. Two or more polypeptide chains interact with each other to form a quaternary structure. Protein folding should be thermodynamically favorable and is supported by hydrophobic interactions, van der Waals forces whereas opposed by conformational entropy.
