The hydrophobic effect is considered to be the major driving force for the folding of proteins having a globular tertiary structure. This effect is observed due to the tendency for hydrophobic molecules to remove themselves from contact with water - hence the tendency of nonpolar substances to aggregate in an aqueous solution and exclude water molecules. 

The association of several protein chains or subunits in a multi-subunit protein is called the quaternary structure of a protein.
