Chapter 17.4, Problem 3CC
Summary Introduction
The recombinant proteins are purified by a common method called polyhistidine tagging or His-tagging. This process involves the addition of a series of histidine residues either to the C- or N- terminus of the protein. The amino acid Histidine is used for tagging because it has a high affinity for the metal ions. Proteins having multiple residues of histidine will preferentially bind to a solid phase material called resin which has exposed to cobalt or nickel atom. This allows the separation of His-tagged proteins from the other cellular constituents.

A recombinant protein corresponding to 52 repeats of the peptide sequence YSPTSPS was covalently linked to agarose beads to make an affinity column. The beads were incubated with a nuclear extract and the protein:beads became phosphorylated. More nuclear extract was run over the column repeatedly and then the beads were washed extensively. The bound proteins were eluted from the column and when they were separated on a SDS-PAGE gel many bands appeared. What cellular process(es) might these proteins be involved in?

- The technique of fluorescence in situ hybridization (FISH) is described. This is another method for examining sequence complexity within a genome. In this method, a DNA sequence, such as a particular gene sequence, can be detected within an intact chromosome by using a DNA probe that is complementary to the sequence. For example, let’s consider the β-globin gene, which isfound on human chromosome 11. A probe complementary to theβ-globin gene binds to that gene and shows up as a brightly colored spot on human chromosome 11. In this way, researchers can detectwhere the β-globin gene is located within a set of chromosomes. Becausethe β-globin gene is unique and because human cells are diploid(i.e., have two copies of each chromosome), a FISH experimentshows two bright spots per cell; the probe binds to each copy ofchromosome 11. A blood sample has been found under the fingernails of the victim, but the sample is a mixture of the victim’s and the killer’s. An SSR screen was run by genetic testing lab to identify which suspect is the killer. The testing lab said that the sequencing machine broke mid-analysis, thus the investigation team only has data from the first 3 SSRs. SSR1 can be either 204 or 188 base pairs long. SSR2 can be either 146 or 122 base pairs long. SSR3 can be either 102 or 88 base pairs long. In the table below, “––––“ means that the allele is present. She was told that a new SNP marker was identified RFPL marker. She requested to do SNP test for all the DNA samples. The results are: her parents are heterozygous for SNPs (that is to say, one chromosome is wild-type and another one contains the SNP). The sister who passed away is homozygous for the SNP. Mary does not have this SNP. So now what is the probability of Mary as a carrier of the death gene?arrow_forward
